Preparation, characterization, and functions of rabbit lymph node cell populations

摘要: Abstract Lymph node cells from rabbits primed with keyhole limpet hemocyanin (KLH) were separated into two populations by passage over goat anti-rabbit Fab′ (AFab′) columns. Cells readily detected surface immunoglobulin (SIg) retained on the column whereas those that passed through lacked SIg and receptors for C3. Adherent cells, eluted purified rabbit IgG or normal serum (NRS) mostly SIg+ enriched complement receptor lymphocytes (CRL) compared to starting, unfractionated population. LNC incorporated much less [ 3 H]thymidine than did non-adherent when stimulated Con A. When induced AFab′, however, only adherent above background levels. Both KLH showed a reduced capacity synthesize anti-KLH antibody, total immunoglobulin, protein in vitro . However, addition of mixture resulted reconstitution anti-KLH, synthesis levels LNC. It was concluded consisted almost exclusively T B cells. By variety criteria suitable many types studies antibody response. not always suited these studies.
