GTPase inhibiting mutations activate the α chain of G s and stimulate adenylyl cyclase in human pituitary tumours

摘要: A subset of growth hormone-secreting human pituitary tumours carries somatic mutations that inhibit GTPase activity a G protein alpha chain, alpha(s). The resulting activation adenylyl cyclase bypasses the cells' normal requirement for trophic hormone. Amino acids substituted in putative gsp oncogene identify domain alpha-chains required intrinsic ability to hydrolyse GTP. This may serve as built-in counter-part separate GTPase-activating proteins GTP hydrolysis by small GTP-binding such p21ras.
