摘要: Normal human melanocytes, which require the phorbol ester 12- O -tetradecanoylphorbol-13-acetate (TPA) for growth in culture, were transfected with a SV40 T-antigen-containing plasmid by using technique of electroporation, and scored colonies morphologically altered cells. The frequency transformed was higher when selection done absence rather than presence TPA medium. Three cell lines derived from characterized. All show an enhanced rate compared to parental cells, anchorage independence, loss dependence on medium supplements growth, chromosomal abnormalities, extended life span, inhibition TPA. They express nerve factor receptor M r 97,000 protein, melanotransferrin, two antigens usually associated melanocytic but cells are not pigmented. three underwent crisis at about passage 10 posttransfection; one line recovered appears have unlimited potential. None is tumorigenic. should be interesting models studying multistage carcinogenesis transcriptional activation
