摘要: Abstract Atg1 is a Serine/Threonine protein kinase that plays pivotal role in autophagy. A complete coding sequence of ATG1 not available for the silkworm, Bombyx mori which good model studying autophagic process. In present study we isolated two full-length cDNAs 2175 (transcript variant A) and 2271 B) bases representing silkworm. Phylogenetic analysis indicated BmATG1 was closely related to orthologs other insects. The encoded BmAtg1 proteins shared extensive homology with from yeast mammals, showing high conservation at N-terminal region where catalytic domain ATP- Mg-binding sites are located. de novo prediction three-dimensional structure each presented. We used real-time RT-PCR quantify dynamic changes mRNA copy number midgut fat body fifth instar larvae undergoing starvation, as well tissues silkworm end last larval instar. Our qPCR results revealed expression levels life were comparable midgut, Malpighian tubules, while these higher gonads; moreover, very different among anterior, middle posterior silk glands. Real-time PCR also showed starvation significantly influenced inducing an upregulation 24 h after food withdrawal, only slight effect midgut. Low observed both control animals up second day spinning phase.
