摘要: Although multiple lasers are now standard equipment on most modern flow cytometers, ultraviolet (UV) (325–365 nm) remain an uncommon excitation source for cytometry. Nd:YVO4 frequency-tripled diode pumped solid-state emitting at 355 nm the primary means of providing UV multilaser cytometers. a number excited fluorochromes available cytometry, cost remains prohibitively high, limiting their use to all but sophisticated instruments. The recent introduction brilliant (BUV) series cell surface marker detection and importance in increasing simultaneous parameters high-dimensional analysis has increased urgency including sources cytometer designs; however, these expensive. Near-UV laser diodes (NUVLDs), direct 370–380 range, have been previously validated cytometric UV-excited probes, quantum nanocrystals, Hoechst dyes, 4′,6-diamidino-2-phenylindole. However, they little-used despite significantly lower cost. In this study, ability NUVLDs excite BUV dyes was assessed, along with compatibility violet (BV) labeling. A NUVLD 375 found least as well source. This slightly longer wavelength did produce some unwanted BV sufficiently low levels require minimal additional compensation. compact, relatively inexpensive that higher power than newest generation small lasers. They can, therefore, make useful, cost-effective substitute traditional multicolor involving dyes.
