摘要: Cytokines are important humoral mediators of beta cell destruction in autoimmune diabetes. The aim this study was to identify novel cytokine-induced genes insulin-producing RINm5F cells, which may contribute death or survival. A global gene expression profile cytokine-exposed cells achieved by automated restriction fragment differential display PCR. selected candidate confirmed real-time RT-PCR analysis. Exposure IL-1β a cytokine mixture (IL-1β, TNF-α, IFN-γ) for 6 h resulted the functional cluster. Apart from well-known up-regulation cytokine-responsive iNOS, NF-κB, MnSOD and Hsp70, several that belong cluster endocytotic pathway were identified. These comprised: clathrin, megalin, synaptotagmin calcineurin, up-regulated mixture. In contrast, calcineurin inhibitor CAIN GDP/GTP exchange protein Rab3 down-regulated cytokines. Other were: agrin, murine adherent macrophage mRNA (MAMA) transport-associated (TAP1/MTP), whereas plasma membrane calcium ATPase (PMCA) 2 PMCA 3 Our results indicate regulated pro-inflammatory This might affect density receptors at surface concomitantly sensitivity toxicity. better understanding cross-talk between signalling pathways could further development strategies protect pancreatic against toxic effects
