摘要: We previously identified a gene, termedMblk-1, that encodes putative transcription factor with two DNA-binding motifs expressed preferentially in the mushroom body of honeybee brain, and its preferred binding sequence, termed Mblk-1-binding element (MBE) (Takeuchi, H., Kage, E., Sawata, M., Kamikouchi, A., Ohashi, K., Ohara, Fujiyuki, T., Kunieda, Sekimizu, Natori, S., Kubo, T. (2001) Insect Mol Biol 10, 487–494; Park, J.-M., Kunieda. Takeuchi, (2002) Biochem. Biophys. Res. Commun. 291, 23–28). In present study, effect Mblk-1 on genes containing MBE Drosophila Schneider's Line 2 cells was examined using luciferase assay. expression transactivated promoters MBEs ∼2–7-fold. Deletion experiments revealed RHF2, second domain Mblk-1, necessary for transcriptional activity. Furthermore, mitogen-activated protein kinase (MAPK) phosphorylated at Ser-444 vitro, Mblk-1-induced transactivation stimulated by phosphorylation Ras/MAPK pathway These results suggest is might function neuronal circuits downstream brain.
