摘要: Publisher Summary The epidermal growth factor (EGF) receptor is a 1186-amino acid transmembrane phosphorylated glycoprotein that mediates the biological effects of EGF. This chapter discusses single-step immunoaffinity chromatography procedure using monoclonal anti-EGF antibody, which competitive inhibitor EGF binding. Desorption with urea gives quantitative recovery denatured protein; elution yields homogeneous active protein. provides convenient single-step, high-yield purification major limitation enzyme eluted EGF, and it not suitable for kinetic or mechanistic analysis activation. However, useful other analyses, such as to understand self-phosphorylation.
