Dihydropyrimidine dehydrogenase activity and thymidylate synthase level are associated with response to 5-fluorouracil in human colorectal cancer.

摘要: In the recent studies associated with modulation of 5-fluorouracil (5-FU) and development new antifolates, attentions have been focused on expression target enzymes, thymidylate synthase (TS) dihydropyrimidine dehydrogenase (DPD), that affect tumor sensitivity resistance to drugs. order evaluate predictability therapeutic efficacy by intratumoral enzyme activity, we investigated role TS content DPD activity in 5-FU. Surgical specimens were obtained from 51 patients colorectal cancer used measure activity. tissues measured [3H]-FdUMP binding assay radioenzymatic assay, respectively. The 5-FU was determined collagen-gel droplet embedded-drug test (CDDST). did not correlate Dukes' staging. There no correlation between any tumors. Simple linear regression analysis showed neither (r=-O.267, p>0.05) nor (r=-O.277, p> 0.05) tumors had a significant effectiveness independently. Four out 24 patients, highly responsive 5-FU, low levels both TS. high value either or proved respond conclusion, these results demonstrate are factors predicting responsiveness cancer.
