摘要: Brain structures such as the outer subventricular zone (OSVZ) and inner fiber layer (IFL) in developing cerebral cortex are especially prominent higher mammals. However, molecular mechanisms underlying formation of OSVZ still largely unknown, mainly because genetic manipulations that can be applied to mammals had been poorly available. Here we developed validated a rapid efficient manipulation technique for germinal zones including using utero electroporation gyrencephalic carnivore ferrets. We also determined optimal conditions express transgenes zones. Using our procedure, morphology GFP-positive cells was clearly visible even without immunostaining, multiple genes were efficiently co-expressed same cells. Furthermore, uncovered fibers, which seemed correspond those IFL monkeys, existed ferrets, derived from newly generated cortical neurons.
