Use of ionic liquid-based extraction for recovery of salmonella typhimurium and Listeria monocytogenes from food matrices.

摘要: Methods for rapid separation (<5 h) and concentration of bacteria based on solubilization complex food matrices have been developed recently to facilitate molecular detection methods. However, a major disadvantage these protocols is the resulting lack viability microorganisms under study due extensive use chemicals enzymes, which can inhibit subsequent quantitative microbiological analyses. By introducing liquid 1-ethyl-3-methylimidazolium thiocyanate into an existing protocol, both methods could be subsequently without losing performance or prolonging analysis. These experiments resulted in average recovery 87% inoculated bacterial cells real-time PCR, 85% nonselective agar plates, 43% selective medium. results illustrate feasibility applying liquids sample pretreatment steps pathogens.
