摘要: Methods for rapid separation (<5 h) and concentration of bacteria based on solubilization complex food matrices have been developed recently to facilitate molecular detection methods. However, a major disadvantage these protocols is the resulting lack viability microorganisms under study due extensive use chemicals enzymes, which can inhibit subsequent quantitative microbiological analyses. In this study, new class organic salts, ionic liquids, were used various foodstuffs, with quantification This approach was applied gram-positive Listeria monocytogenes gram-negative Salmonella enterica serovar Typhimurium. By introducing liquid 1-ethyl-3-methylimidazolium thiocyanate into an existing protocol, both methods could be subsequently without losing performance or prolonging analysis. These experiments resulted in average recovery 87% inoculated bacterial cells real-time PCR, 85% nonselective agar plates, 43% selective medium. results illustrate feasibility applying liquids sample pretreatment steps pathogens.
