摘要: To identify DNA sequences of the Arabidopsis thaliana chalcone synthase gene (CHS) concerned with induction by UV-B and UV-A/blue light, AtCHS promoter constructions were assayed transient expression in protoplasts prepared from two different lines cultured A. cells. The responded similarly to leaf tissue light-dependent CHS transcript accumulation. reporter enzyme beta-glucuronidase (GUS) was used monitor light-responsive activity. A 1972 bp conferred light GUS Deletion 164 resulted reduced strength but retention responsiveness light. Further deletion abolished transcriptional contains closely resembling LRUPcCHS, (light-responsive unit Petroselinum crispum promoter). This region, designated LRUAtCHS, sufficient confer a heterologous core promoter. Mutation LRUAtCHS corresponding ACGT element MYB recognition LRUPcCHS inactivation 335 deletions. However, mutant 668 retained residual light-induced expression, indicating presence additional functional upstream -335. single G-box-like sequence around -442 had no effect on responsiveness, that it does not function regulation this Since difference observed any variant, we conclude phototransduction pathways regulate transcription factors which interact common elements. results from-our analysis will facilitate study genetic means thaliana.
