摘要: A fusion protein containing the full-length sequences of mitogen, basic fibroblast growth factor (FGF-2), and ribosome-inactivating protein, saporin (SAP), has been expressed in E. coli. As expected, it binds with high affinity to heparin-Sepharose like FGF-2 can displace binding radiolabeled its receptor. In contrast, only much lower activity than free saporin, although full be generated by proteolytic removal moiety. Cytotoxicity experiments B16-F10 mouse melanoma cells establish that is active as a chemical conjugate against these intact cells. Presumably have ability activate SAP component through an intra-cellular metabolism protein. Because we also show tumor inhibition properties antimetastatic vivo models melanoma, findings support hypothesis FGF-based ligand-mediated cytotoxicity serve target cytotoxic agents vivo.
