摘要: The fibrinolytic enzyme cascade is a summary term for several regulatory serine proteases and protease inhibitors, which cooperate in the digestion of extracellular matrix protein processes tissue repair, growth, remodelling. Tissue-type plasminogen activator (t-PA) urinary-type (u-PA) activate proenzyme by cleavage single peptide bond, converting it into plasmin, proteolytic with specificity similar to pancreatic digestive trypsin. Plasminogen synthesized liver secreted bloodstream where circulates relatively large amounts (80160 mg/l). Blood plasma provides reservoir activity recruited removal fibrin deposits or proteins during morphogenesis, wound healing malignant growth. t-PA u-PA are structurally enzymatically related. Both display characteristic mosaic molecular architecture: they consist series structural motifs homologous other proteins. Thus, starting from amino terminus, composed “finger” domain, found tandemly arranged fibronectin; an epidermal growth factor (EGF) like motif, likewise various blood clotting factors, receptor developmentally proteins; two “kringle” regions, present u-PA, plasminogen, prothrombin, XII apolipoprotein (a) finally sequence trypsin, chymotrypsin, elastase kallikrein. “finger”- one “kringle”-domains absent u-PA.
