DNA Replication-Coupled PCNA Mono-Ubiquitination and Polymerase Switching in a Human In Vitro System

摘要: Abstract Translesion DNA synthesis is a mechanism of damage tolerance, and mono-ubiquitination proliferating cell nuclear antigen (PCNA) considered to play key role in regulating the switch from replicative translesion polymerases (pols). In this study, we analyzed effects pol δ on PCNA with ubiquitin-conjugating enzyme ligase UBE2A/HHR6A/RAD6A–RAD18. The results revealed that interacting better target for ubiquitination, could be coupled replication. Consequently, reconstitute replication-coupled switching between pol, η, an ultraviolet-light-irradiated template. With system, obtained direct evidence polymerase reactions are stimulated by PCNA, depending function ubiquitin binding zinc finger domain η. This study provides framework detailed analyses molecular mechanisms human regulation synthesis.
