Diminishing Efficacy of Octreotide (SMS 201-995) on Gastric Functions of Healthy Subjects During One-Week Administration

摘要: In this study, the pharmacokinetics, efficacy, and tolerability of 25 100 micrograms octreotide given t.i.d. for 7 days subcutaneously were investigated in 12 healthy male subjects. Serum concentrations drug well reproducible within 1 wk. Octreotide significantly raised 24-h median intragastric pH on day 1, but no longer 6. Peptone-stimulated gastric acid volume secretion markedly less suppressed by compared with 2. gastrin release was abolished 2 7, as peptone-stimulated insulin release. Blood glucose altered a biphasic pattern 7. All effects without clear-cut dose-response relationship. A mean half-life 115 min calculated. Dose-unrelated side (e.g., abdominal cramps, diarrhea, fatty stools) registered. conclusion, is powerful inhibitor during acute treatment. Its loss efficacy 1-wk administration may be due to adaptation somatostatin receptors hormonal counterregulation.
