Effects of a finite-sized detector on the OTF of confocal fluorescent microscopy

摘要: For a confocal fluorescent microscope with finite sized circular detector, the 3-D optical transfer function (OTF) for thick object has been developed without using Stockseth's approximation, and in-focus (2-D) OTF thin also derived. Both OTFs were numerically calculated to reveal dependence on radius of detector. The results show that have negative values when detector exceeds certain magnitudes approach those conventional incoherent as becomes infinite
