摘要: Abstract Intact reticulocytes incubated in vitro synthesize hemoglobin at a linear rate for 1 h 30°. During the first 30 min about one-third of 80-S ribosomes are transferred into polysome fraction, leading to an increase concentration and size polysomes. Protein synthesis is accompanied by breakdown, this dominant event. These changes ribosome distribution appear be result initiation process (attachment polysomes) rather than rates peptide bond formation or release completed polypeptide chains from ribosomes. The rate-limiting step protein was located by: (a) determining synthetic time β-chain within two different intervals during incubation, (b) investigating kinetics under conditions where amino acids were limiting.
