Pharmacokinetic parameters of (R)-(-) and (S)-(+)-flurbiprofen in dairy bovines.

摘要: The aim of the study was to evaluate pharmacokinetics flurbiprofen (FBP) in different age groups and physiological status dairy cattle. Ten Argentine Holstein bovines were divided into three groups: 3 cows early lactation, gestation 4 newborn calves. Based on previous experience, all animals received racemic FBP (50:50) at a dose 0.5 mg/kg by intravenous administration. Blood samples taken predetermined times after administration flurbiprofen. Plasma enantiomer concentrations measured HPLC. Total body clearance (ClB) (S)-(+)-FBP higher calves than (114.5, 136.4, 121.4, 128.9 μg/ml vs 22.0, 24.2, 46.5 27.6, 25.3, 34.6 μg/ml). In disposition kinetics showed stereoselective behaviour. Area under concentration–time curve (AUC) ClB steady-state volume distribution (V ss) lower for (R)-(−)-FBP (S)-(+)-FBP. cows, stereoselectivity observed elimination half-life $$(frac{t_1}{2})$$ only lactation group. this study, enantioselective metabolic behaviour situations studied found. Hence, it is possible that both enantiomers may contribute drug's therapeutic effects, but further studies with separate will be required elucidate their metabolism.
