Induction of virus multiplication in 3T3 cells transformed by a thermosensitive mutant of polyoma virus: II. Formation of oligomeric polyoma DNA molecules

摘要: Abstract 3T3 cells transformed with a thermosensitive mutant of polyoma ( Ts-a ) can be induced to produce virus if the (Ts-a-3T3) are transferred from 38.5 °C 31 (Vogt, 1970). Viral DNA synthesis is detectable in activated cell population 24 hours after shift low temperature and steadily increases for several days. Following activation at °C, cultures continue make viral least they shifted back high temperature. Our data suggest that induction multiplication involves asynchronous occurrence unique event, which even under non-permissive conditions. Superhelical closed circular dimers trimers synthesized as well monomeric molecules. These oligomers amount 40% total DNA; not obligatory precursors form, only type found mature virions. Oligomers observed (1% or less) infection either wild polyoma. Several possible models explain origin oligomers, state nature activating event discussed.
