TEM samples have be thin and examined vacuum; therefore only solid can investigated. most common popular way prepare for subject them chemical fixation, staining, dehydration, embedding resin (all these steps introduce considerable artifacts) before investigation. An alternative immobilize by cooling. High pressure freezing so far approach vitrify (water solidification without ice crystal formation) bulk about 200 micrometer thick. This method leads an improved ultrastructural preservation. After high freezing, subjected follow-up procedure, such freeze-substitution embedding. also sectioned into frozen hydrated sections analyzed cryo-TEM. Also immunocytochemistry, good practicable way.
