摘要: A light plasma membrane fraction corresponding to a buoyant density of 1.087 ± 0.005 g/cm3 and devoid chlorophyll was prepared purified from Anacystisnidulans according recently published procedure (G.A.Peschek, V.Molitor, M.Trnka, M.Wastyn W.Erber (1988) Methods Enzymol. 167, 437–449). Besides major amounts carotenoids the membranes contained small but significant pool chlorophyllide protochlorophyllide as verified by room temperature 77K spectrofluorimetry analytical separation identification high performance liquid chromatography using authentic standards. Incubation in strict darkness presence NADPH accompanied gradual stoichiometric replacement chlorophyllide, NADP+ effecting reverse transition. The reaction completely insensitive illumination (5–20 w/m2 tungsten light) abolished after heating (90°C, 5 min) or 10 mM EGTA, specifically stimulated calcium ions. Our results indicate occurrence light-independent NADPH: oxidoreductase activity Anacystisnidulans.
