摘要: In spite of previous reports, the precise role bone morphogenetic proteins (BMPs) on cardiomyocyte differentiation, especially in absence or presence minimum amount serum culture medium is still unclear. So, aim present study was to investigate effect BMP-4 mouse embryonic stem cells (ESCs)-derived differentiation serum-free and low-serum media. The ESCs cardiomyocytes induced by embryoid bodies' (EBs') development through hanging drop, suspension plating stages. Different models were designed according addition fetal bovine (FBS) knockout™ replacement (KoSR) three 10 ng/ml added throughout period. Up 30 days after plating, contraction beating frequency monitored evaluated daily. growth characteristics assessed cardioactive drugs, immunocytochemistry, transmission electron microscopy (TEM) reverse transcription-polymerase chain reaction (RT-PCR). complete serum, neither control nor treated groups resulted cardiac differentiation. Addition FBS drop stage appearance clusters some EBs. best model which only first 24 h carried out at FBS, treatment EBs characterized positive immunostaining for applied antibodies, chronotropic response drugs cardiac-specific genes expression different developmental These showed immature myofibrils numerous intercellular junctions. conclusion, unable induce from models, least small necessary. Furthermore, factors are not strictly necessary initial activation, but they do favor a better cardiomyocytes.
