Gene Expression Analysis of Whole Blood, Peripheral Blood Mononuclear Cells, and Lymphoblastoid Cell Lines From the Framingham Heart Study

摘要: Despite a growing number of reports gene expression analysis from blood-derived RNA sources, there have been few systematic comparisons various sources in transcriptomic or for biomarker discovery the context cardiovascular disease (CVD). As pilot study Systems Approach to Biomarker Research (SABRe) CVD Initiative, this investigation used Affymetrix Exon arrays characterize three sources: lymphoblastoid cell lines (LCL), whole blood using PAXgene tubes (PAX), and peripheral mononuclear cells (PBMC). Their performance was compared relation identifying transcript associations with sex risk factors, such as age, high-density lipoprotein, smoking status, differential count. We also identified set exons that vary substantially between participants, but consistently each source. Such are thus stable phenotypes participant may potentially become useful fingerprinting biomarkers. In agreement previous studies, we found is distinct. Unlike PAX PBMC, LCL showed little association LCL, however, able detect two genes related status. PBMC Y-chromosome probe sets similarly slightly better than LCL.
