Light elements quantification in stimulated cells cryosections studied by electron probe microanalysis.

摘要: The quantification of intracellular light elements such as carbon, nitrogen and oxygen may be useful in understanding the biological mechanisms, for example generation nitric oxide which is involved apoptosis inflammatory reaction. Electron energy loss spectroscopy (EELS) coupled with scanning transmission electron microscopy a method to detect thin cryosections. Recent developments X-ray detectors ultra-thin protection windows allows detection by dispersive spectroscopy. In present study, we have demonstrated using both methods that stimulation BV-2 murine microglial cell line γ-interferon lipopolysaccharide leads increase concentration no change concentration. This indicates use exogeneous molecular response stimulation. But detected could not only due NO expression because production was 1000 times less than observed.
