Oxygen radicals and nitric oxide in rat mesenteric ischaemia-reperfusion: modulation by L-arginine and NG-nitro-L-arginine methyl ester.

The aims of the present study were to detect changes in superoxide anion (O2.-), nitric oxide (NO) and other reactive oxygen species (ROS) directly by measurement chemiluminescence (CL) investigate role L-arginine, a synthase (NOS) substrate, NG-nitro-L-arginine methyl ester (L-NAME), NOS inhibitor, together with their molecular enantiomers D-arginine D-NAME, rat mesenteric ischaemia-reperfusion (I/R) model. 2. Seventy-nine female Wistar albino rats divided into eight groups. first three groups underwent sham operation; group 1 was control group, 2 received L-arginine 3 L-NAME. Ischaemia produced remaining five ligation superior artery for 30 min followed 60 reperfusion. Group 4 I/R 5-8 either L-NAME, or respectively. 3. Both luminol lucigenin CL significantly increased compared sham-operated L-Arginine reduced measurements. D-Arginine also protective, but not as much L-arginine. L- had vitro O2.- (-)scavenging potential, tested xanthine-xanthine oxidase system. NG-Nitro-L-arginine decreased lipid peroxidation values addition reducing Nitric concentrations comparison Peroxynitrite formation I/R. Treatment L-NAME beneficial NO reperfused ileum. 4. In our model, O2.-, ROS increased. Although inhibitors effective oxidative damage, increasing beneficial, presumably due ability inhibit phagocyte adherence its radical scavenging potential. fact, may have different effects terms tissue injury protection depending on concentration haemodynamic state tissue.
