摘要: Genome integrity is maintained, despite constant assault on DNA, due to the action of a variety DNA repair pathways. Nucleotide excision (NER) protects genome from deleterious effects UV irradiation as well other agents that induce chemical changes in bases. The mechanistic steps required for eukaryotic NER involve concerted at least six proteins or protein complexes. specificity incise only strand including damage defined positions determined by coordinated assembly active complexes onto damaged DNA. In order understand molecular mechanism reactions and origin this control we analyzed architecture functional nanometer resolution scanning force microscopy (SFM). initial step recognition XPC-HR23B observe induced change conformation. induces bend upon binding stabilized site damage. We discuss importance XPC-HR23B-induced distortion an architectural feature can be exploited subsequent complex.
