摘要: The c-Myb protein is a transcriptional regulator initially identified by homology to the v-Myb oncoprotein, and has since been implicated in human cancer. most highly conserved portion of DNA-binding domain which consists three imperfect repeats. Many other proteins contain one or more Myb-related domains, including number that do not bind directly DNA. We performed phylogenetic analysis diverse classes domains discovered patch acidic residues common all domains. These are positioned first alpha-helices within each repeats comprise domain. Interestingly, these present on surface distinct from binds Alanine mutagenesis revealed third repeat essential for activity, but neither nuclear localization nor DNA-binding. Instead, required efficient chromatin binding interaction with histone H4 N-terminal tail.
