Canine leptin: cDNA cloning, expression and activity of recombinant protein.

摘要: Abstract Leptin, the product of ob gene, is one key molecules for regulation appetite and whole-body energy balance, thereby pathogenesis obesity. In an attempt to clarify roles leptin in obesity and/or related diseases companion animals, canine c DNA was cloned by amplifying reverse-transcriptase products RNA extracted from adipose tissue beagle. A clone about 3 kbp contained a 501 bp open reading frame coding 167-amino acid protein with 21-amino signal peptide. The sequence 146-amino mature more than 79 per cent identical those other mammals. Northern blot analysis revealed abundant expression m tissue, but not tissues, adult beagles. When Chinese hamster ovary cells expressing rat receptor were stimulated recombinant produced E. coli , some intracellular transduction proteins phosphorylated, indicating that biologically active. data reported herein will be helpful further studies dog health disease.
