Hypoxia-mediated downregulation of miRNA biogenesis promotes tumour progression

摘要: Cancer-related deregulation of miRNA biogenesis has been suggested, but the underlying mechanisms remain elusive. Here we report a previously unrecognized effect hypoxia in downregulation Drosha and Dicer cancer cells that leads to dysregulation increased tumour progression. We show hypoxia-mediated is dependent on ETS1/ELK1 transcription factors. Moreover, mature array deep sequencing studies reveal altered maturation under hypoxic conditions. At functional level, this phenomenon results progression vitro vivo, data from patient samples are suggestive tumours. Rescue by siRNAs targeting vivo significant regression. These findings provide new link mechanistic understanding global microenvironment.
