摘要: The complete RNA genome of the avian nephritis virus (ANV) associated with acute in chickens has been molecularly cloned and sequenced. Excluding poly(A) tail, comprises 6,927 nucleotides contains three sequential open reading frames (ORFs). first ORF (ORF 1a) a sequence encoding serine protease motif, second 1b) an RNA-dependent polymerase. 1a may be linked to by ribosomal frameshifting mechanism. third 2) encode virion structural proteins as polyprotein precursor. Two RNAs, probably genonic subgenonic (7.5 3.0 kb), were detected cytoplasm ANV-infected cells. ANV human astroviruses have same organization, both are characterized presence two bands. amino acid homologies products 1a, 1b, 2 20.3, 41.9, 25.8% corresponding ORFs astrovirus serotype 1 (A/88 Newcastle strain). We constructed genonic-length cDNA clone test whether vitro transcript is infectious. When chicken kidney cell culture was transfected transcribed clone, infectious produced cytopathic effects absence trypsin. These observations suggested that (G-4260 strain) new genus family Astroviridae.
