Morphological, molecular and MALDI-TOF mass spectrometry identification of ixodid tick species collected in Oromia, Ethiopia.

摘要: Matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) technology has recently been reported as a promising method for arthropods identification. More recently, our laboratory the correct identification of tick species via MALDI-TOF MS protein spectra profiling legs from fresh specimens. The aim present study was to assess use ixodid preserved in 70 % ethanol during field collection Ethiopia. Following morphological 12 species, 85 specimens were subjected MS. Spectral analysis revealed an intra-species reproducibility and inter-species specificity that consistent with classification. To support results identification, 41 comprising 3 5 per used create reference database, which evaluated using 44 remaining blind tests 100 % studied by correctly identified. A relevant Log score value (LSV) >1.8 recorded all MS, except Rhipicephalus praetextatus. identifications confirmed sequencing 12S ribosomal RNA (rRNA) gene 40 belonging 11 species. Taken together, indicate is reliable tool even after preservation ethanol, provided database built represent respective stored under same conditions.
