Molecular characterization and epigenetic regulation of Mei1 in cattle and cattle-yak.

摘要: Mei1 is required for the homologous recombination of meiosis during mammalian spermatogenesis. However, knowledge about bovine (bMei1) still limited. In present study, we cloned and characterized bMei1, investigated epigenetic regulatory mechanism bMei1 expression in vivo vitro. The full length coding region was 3819bp, which encoded a polypeptide 1272 amino acids. Real-time PCR showed that mRNA level testis cattle-yak with meiotic arrest male infertility significantly decreased as compared cattle (P<0.01). Conversely, methylation levels promoter gene body were increased. Additionally, mammary epithelial cells (BMECs) activated by treatment methyltransferase inhibitor 5-aza-2' deoxycytidine (5-Aza-CdR). Our data suggest may play an important role spermatogenesis be involved sterility, its transcription regulated DNA methylation.
