TP53 mutations and MDM2 gene amplification in squamous-cell carcinomas of the esophagus in south Thailand.

摘要: Squamous-cell carcinoma of the esophagus (SCCE) shows geographic variations in incidence that are thought to reflect etiological involvement environmental or dietary risk factors. Mutations TP53 frequent SCCE, and there is evidence both frequency type these mutations may differ from one area other. Although SCCE relatively rare most parts Thailand, province Songkhla (south Thailand) has been described as a high-risk for SCCE. We have analyzed 56 cases this by denaturing gradient gel electrophoresis (DGGE, exons 5-8) direct DNA sequencing. The same tumors were also MDM2 gene amplification differential PCR. detected 23 (41%). In contrast, clear was only 2 (4%), which contained wild-type TP53. Comparison with published results other areas high revealed spectrum south Thailand similar observed central China (Henan Province) but clearly differs western Europe (Normandy, France; northern Italy), more G:T transversions fewer affecting A T base pairs.
