摘要: The p53 tumor suppressor gene is frequently mutated and the K- ras oncogene occasionally in primary specimens of human lung carcinomas. These genes also cooperate immortalization neoplastic transformation rodent cells. To determine whether these mutations are necessary for maintenance immortalized and/or neoplastically transformed states bronchial epithelial cells, regions (K-, H-, N-) were sequenced nine carcinoma cell lines. Detection by polymerase chain amplification direct DNA sequencing was corroborated immunocytochemistry coimmunoprecipitation with heat shock protein 70. frequently, but not always, data consistent hypothesis that multiple genetic changes involving both protooncogenes occur during carcinogenesis.
