摘要: AIMS--Tissue fibrosis is a common and serious consequence of chronic inflammation. The mechanism linking these two processes poorly understood. present study has utilised human in vivo model delayed type hypersensitivity (DTH) reaction, the tuberculin Heaf induced by intradermal BCG immunised subjects, to dissect relation between processes. METHODS--Punch skin biopsy specimens were obtained on day 5, 13 six 16 weeks following test 18 subjects with grade 3 or 4 responses. Skin from served as controls. examined using immunohistochemical staining for 1 procollagen transforming growth factor-beta (TGF-beta), well situ hybridisation messenger RNA (mRNA). RESULTS--Immunohistochemical analysis revealed increased deposition TGF-beta tissue matrix 5 test. There was also extensive early 5. Procollagen-1 maximal 13, sites up eight after inoculation. localised within cells surrounding areas inflammatory infiltrate perivascular tissues. presence new collagen formation confirmed oligonucleotide probes mRNA sections 13. CONCLUSIONS--These data DTH response indicate that immune intimately associated an increase production factors initiation fibrotic response.
