摘要: Abstract A novel saponin (JS) was isolated from the tetraploid jiaogulan, a popular tea also known as Gynostemma pentaphyllum , and characterized for its chemical structure. JS investigated inhibitory effect on adipocyte differentiation potential molecular mechanisms using 3T3-L1 murine pre-adipocytes. treatment down-regulated expression of peroxisome proliferator-activated receptor γ (PPARγ) CCAAT/enhancer-binding protein α (C/EBPα) at both mRNA levels. Further analysis proved that inhibition primarily limited to early stage differentiation, where decreased DNA-binding activity C/EBPβ by blocking phosphorylation. In addition, increased phosphorylation glycogen synthase kinase 3β (GSK-3β) subsequently stability nuclear translocation β-catenin through modulating Wnt/β-catenin signaling components. Moreover, JS-induced down regulation adipogenic markers fat accumulation is β-catenin-dependent. These data suggested this compound might inhibit adipogenesis activating during differentiation.
