Enhancement of lipopolysaccharide-induced toll-like receptor 2 expression and inflammatory cytokine secretion in HUVECs under high glucose conditions

摘要: Abstract Aims Endothelial inflammatory responses mediated by toll-like receptors (TLRs) play an important role in atherogenesis. We aimed to investigate the exacerbation of response human umbilical vein endothelial cells (HUVECs) under high glucose conditions. Main methods HUVECs were exposed normal (5.5 mmol/L) and (25, 50 mmol/L), alone or with lipopolysaccharide (LPS 0, 10, 100, 1000 μg/L). Then concentrations TNF-α IL-6 culture supernatants determined. The expression receptor 2 (TLR2), TLR4 NF-κB was evaluated Western blot RT-PCR analysis. Key findings Compared group, exposure 50 mmol/L 1000 μg/L LPS significantly increased supernatants. Neither 25 mmol/L nor low concentration (≤ 100 μg/L) had effect on release, TLR2 expression, but they stimulated conditions (25 mmol/L) combination. (100 μg/L) did not alter condition.
