摘要: Both cell polarity and c-Jun N-terminal kinase (JNK) activity are essential to the maintenance of tissue homeostasis, disruption either is commonly seen in cancer progression. Despite established connection between loss-of-cell JNK activation, much less known about molecular mechanism by which aberrant induces JNK-mediated migration tumor invasion. Here we show results from a genetic screen using an vivo invasion model via knocking down gene Drosophila wing discs, identify Rho1-Wnd signaling as important link that mediates polarity-triggered activation We Wallenda (Wnd), protein mitogen-activated family, forming complex with GTPase Rho1, both necessary sufficient for Rho1-induced JNK-dependent invasion, MMP1 epithelial-mesenchymal transition. Furthermore, Wnd promotes proliferation growth through wingless production when apoptosis inhibited p35. Finally, shows oncogenic cooperation Ras(V12) trigger eye discs causes into ventral nerve cord. Together, our data not only provides novel mechanistic insight on how loss contributes but also highlights value system explore human biology.
