摘要: PURPOSE. To investigate the effect of epidermal growth factor (EGF) on induction phosphatidylinositol 3-kinase (PI 3‐ kinase) gene expression during rabbit corneal epithelial wound repair. METHODS. Epithelial wounds (6 mm in size) were created corneas and EGF (2 mg) applied every 8 hours to one eye, other eye served as a control. The repair was monitored by staining tissue with fluorescein followed photography. area quantified computer program. At different time intervals, rabbits killed epithelium used for estimation PI kinase activity, western blot analysis, or reverse transcription‐polymerase chain reaction (RT‐PCR). For situ hybridization, whole sectioned sections processed mRNA probes. RESULTS. In untreated progressively healed time-dependent manner, 75% closed at 48 post wounding. Application further stimulated all completely hours. Analysis showed increase its enzyme activity that maximally increased 36 hours, when nearly closed. Western analysis revealed amounts protein course RT‐PCR products from tissues, taken times repair, maximum A visible also detected hybridization This CONCLUSIONS. results indicate temporal correlation between activation Topical application stimulates kinase. It is suggested may play role EGF-induced cell proliferation (Invest Ophthalmol Vis Sci. 1999;40:2819 ‐2826)
