Increased angiogenic capabilities of endothelial cells from microvessels of malignant human gliomas

摘要: Vascular endothelial cells (ECs) that initiate tumor angiogenesis may acquire distinct properties after conditioning in microenvironment as compared to ECs non-malignant tissues. Thus far, most vitro studies of used isolated from normal tissues, which not fully represent the nature vasculature. In this study, glioma-derived microvascular (GDMEC) were purified human glioma tissues by incubating with magnetic beads coated anti-CD105 antibody and highly pure (98%) preparations GDMEC obtained. These exhibited typical EC phenotype, proliferated rapidly culture. Interestingly, expressed higher levels VEGF receptors, flt-1 flk-1, an established cell line ECV304 primary umbilical vascular (HUVEC). Functionally, capable forming intercellular junctions tubule-like structures (TLS) various sizes. Stimulation further promoted TLS formation diverse tubular walls GDMEC. contrast, formed HUVEC showed significantly different features. We observed Nordy, a synthetic lipoxygenase inhibitor, potently inhibited The results suggest isolation derived is more appropriate for test potential anti-cancer therapeutic targets.
