Polysaccharides from Ganoderma formosanum function as a Th1 adjuvant and stimulate cytotoxic T cell response in vivo.

摘要: The fungus of Ganoderma is a basidiomycete that possesses variety pharmacological effects and has been used in traditional Asian medicine for centuries. formosanum native species isolated Taiwan, we have previously demonstrated PS-F2, polysaccharide fraction purified from the submerged culture broth G. formosanum, exhibits immunostimulatory properties macrophages. In this study, further characterized adjuvant functions PS-F2. vitro, PS-F2 stimulated dendritic cells (DCs) to produce proinflammatory cytokines, including TNF-α, interleukin (IL)-6, IL-12/IL-23 p40. also DCs express maturation markers CD40, CD80, CD86, MHC class II. murine splenocyte culture, treatment resulted elevated expression T-bet interferon (IFN)-γ T lymphocytes. When as an vivo with ovalbumin (OVA) antigen, OVA-specific antibody production primed IFN-γ PS-F2-adjuvated immunization induced CTLs, which protected mice challenge tumor expressing OVA. Collectively, our data show capable inducing Th1-polarized adaptive immune response, would be useful vaccines against viruses tumors.
