Red blood cell storage in SAGM and AS3: a comparison through the membrane two-dimensional electrophoresis proteome.

摘要: Background. SAGM is currently the standard additive solution used in Europe, while AS-3 third that has been licensed USA, and also one part of Canada. Although based on a saline-adenine-glucose solution, it contains citrate phosphate. Storage red blood cell concentrates CPD-SAGM known to lead accumulation wide series storage lesions, including membrane protein fragmentation vesiculation, as we could previously determine through 2-dimensional gel electrophoresis. Materials methods. Through 2D-SDS-IEF-polyacrilamide electrophoresis performed time course analysis (day 0, 21 42 storage) membranes from leukocyte-filtered either stored or CP2D-AS-3. Results discussion. From present study emerges profile cells presence appears be slightly different (better than) previous reports SAGM-stored counterparts. However, increase total spot number due fragments at day significant decrease are suggestive universal phenomenon which not efficiently tackled by two solutions investigated study. Conclusion. To further delve into lesion issue for RBCs AS-3, would interesting future assay metabolic changes over progression well.
