摘要: A method is described for the simultaneous analysis of S-adenosylmethionine (SAM) and its metabolites, S-adenosylhomocysteine (SAH) decarboxylated along with natural polyamines, putrescine, spermidine spermine. The separation obtained by a reversed-phase ion-pair liquid chromatographic procedure gradient elution followed dual detection. UV absorbance at 254 nm used SAM whereas polyamines some major amino acids, e.g., methionine, tyrosine tryptophan, are analyzed fluorescence detection after UV-cell derivatization o-phthalaldehyde. separate high-performance (HPLC) using isocratic electrochemical employed to analyse in same tissue extracts catechols 5-hydroxyindoles, 3,4-dihydroxyphenylalanine (DOPA), dopamine, norepinephrine, 3,4-dihydroxyphenylacetic acid, homovanillic 4-hydroxy-3- methoxyphenylalanine , 5-hydroxytryptophan, serotonin 5- hydroxyindolacetic acid. sample preparation two HPLC procedures requires only homogenization tissues perchloric acid centrifugation before injection onto column. have been applied study interrelationship, various rats, between SAH levels biogenic different treatments L-DOPA alone or combination alpha- monofluoromethyl -DOPA, potent enzyme-activated irreversible inhibitor aromatic L-amino decarboxylase.
