摘要: Glucose is an important metabolite and its presence during in vitro oocyte maturation (IVM) can have profound effects on the oocyte's developmental capacity. We demonstrated that glucose uptake increases over a 24 h IVM period, with most accounted for as l-lactate production. However, proceeds, production remains constant, suggesting alternative role metabolism. hypothesised latter stages of maturation, not by utilised FSH-stimulated extracellular matrix (ECM) synthesis. To examine precursor utilisation synthesis ECM, bovine cumulus-oocyte complexes (COCs) were matured +/- FSH and/or glucosamine (an substrate components). Measurements included COC diameters, consumption spent media [U-(14)C]glucose incorporation into ECM. significantly stimulated both diameter 20-24 compared unstimulated complexes, although co-incubation decreased total 1.7-fold alone (P < 0.05). Furthermore, there was linear relationship between metabolism glucosamine, majority being via glycolysis. In twofold less incorporated COCs cultured without glucosamine. These results support hypothesis link ECM has multiple fates within levels are dependent composition environment.
