Are ancient durum wheats less toxic to celiac patients? A study of α-gliadin from Graziella Ra and Kamut.

摘要: In the present paper, controversial hypothesis suggesting ancient grains might show lower immunogenic properties and therefore possibility to introduce them in diet of wheat-sensitive people, including celiac patients, was investigated. The potential durum wheats Graziella Ra Kamut studied by comparison accessions Cappelli, Flaminio, Grazia Svevo. Experiments were carried out with two monoclonal antibodies (mAbs) raised against α-gliadin peptides p31–49 p56–75 (the latter containing overlapping DQ2-Glia-α1 DQ2-Glia-α2 epitopes), toxic for patients. For all accessions, a few alleles also cloned, sequenced translated into aminoacid sequences. Several substitutions or deletions detected p31–49, epitopes, nevertheless, ELISA constantly showed antibody-antigen positive reactions which led us suggest that mAbs binding not apparently affected polymorphisms. Moreover, observed DQ2-Glia-α3 DQ8-Glia-α1 epitopes. Although some variants evidenced herein previously reported have diminished abolished T cell stimulatory capacity, results cannot confirm would be less CD-toxic. conclusion, we strongly advice patients from consuming Kamut.
