Biochemical and immunochemical characterization of the antigen–antibody reaction on a non-toxic biomimetic interface immobilized red blood cells of crucian carp and gold nanoparticles

摘要: A special protein assay system based on a highly hydrophilic, non-toxic and conductive biominetic interface has been demonstrated. To fabricate such system, red blood cells of crucian carp (RBC) was initially grown glassy carbon electrode surface (GCE) deposited nano-sized gold particles (GPs), second nanoparticle layer (NG) then absorbed the RBC surface, finally mammary cancer 15-3 antibody (anti-CA15-3) attached functional surface. competitive immunoassay format employed to detect CA15-3 with horseradish peroxidase (HRP)-labeled as tracer hydrogen peroxide enzyme substrate. When immunosensor incubated into mixture solution containing HRP-labeled sample for 1h at 37 degrees C, amperometric response decreased increment concentration. AFM images modified revealed uniform distribution nanogold. In situ QCM electrochemical measurements demonstrated that wanted antibody-antigen reactions should occur high specificity selectivity. The specific can be developed further yield sophisticated structures other proteins.
