Down regulation of epidermal growth factor receptors: direct demonstration of receptor degradation in human fibroblasts.

摘要: The metabolism of the receptor for epidermal growth factor (EGF) has been measured by labeling in vivo with radioactive amino acid precursors and then determining, immunoprecipitation specific anti-EGF antisera, rate degradation when cells are placed a nonradioactive medium. In human fibroblasts EGF (t1/2 = 10.1 h) was faster than total cell protein. When added to medium, half-life prelabeled decreased 1.2 h fibroblasts. These data demonstrate direct analysis protein that during "down regulation" is rapidly degraded. Enhanced observed 5-10 min after addition EGF. EGF-induced blocked methylamine, chloroquine, iodoacetate, or incubation at 25 degrees C.
