Human osteoclast formation and activity on an equine spongy bone substitute

摘要: OBJECTIVES The aim of the present study was to evaluate in vitro formation and activity human osteoclasts (OCLs) generated on a new type xenograft for bone substitution, an equine spongy bone. MATERIAL AND METHODS Peripheral blood mononuclear cells from healthy volunteers were used generate OCLs presence macrophage colony stimulating factor (M-CSF) receptor activator NF-kappaB ligand (RANKL) bovine slices (positive control) Morphological biochemical methods assess activity. RESULTS Cells after 21 days culture showed similar morphology those positive control displayed typical OCL markers features, indicating that this material supported formation. Moreover, these functionally active with statistically significant differences compared release tartrate-resistant acid phosphatase (TRAcP5b) at 14 culture. With regard resorption, bone, formed smaller discontinuous island-like lacunae rather than lobulated, tracking resorption observed control. CONCLUSIONS This enables clinicians tailor usage presents model, which can be applied preclinical assessment substitute material's resorbability rates.
