摘要: Leukemic cells from ten patients with "nonsecretory," B-type chronic lymphocytic leukemia (CLL) were cultured alone or in the presence of Epstein-Barr virus (EBV) a phorbol ester (12-0-tetradecanoylphorbol-13-acetate; TPA) for 7 days. At periodic intervals cell morphology, cytoplasmic immunoglobulin content (direct immunofluorescence) and capacity secretion (hemolytic plaque assay) assessed. A variable but significant number TPA-treated CLL all expressed single light-chain type at some stage, usually within first 3 EBV induced similar changes seven eight cases tested. Untreated cultures negative contained few immunoglobulin-positive cells. Cells five nine secreted type. In every instance this was identical to surface immunoglobulin. Cytologic observed leukemic after treatment one both agents cases. The major feature an increase size associated immunoblastic plasma-cytoid features. Mitotic figures binucleate also present. These studies indicate that TPA are effective inducing synthesis "nonsecretory" B neoplasms useful tools studying maturation potential inherent these tumors. study suggests is monoclonal product.
